I'm in the Columbia area and have been all over the state pretty much.  The Palmetto Trail is probably the best longer route.  It goes from the NW corner all the way across to Charleston.  There are some routes that can be put together for 50-100 miles that cover a lot of back/dirt roads but nothing aimed at bikepacking really established, at least around me.  Taking a trip up to NC offers a few excellent routes.
